响应面优化连翘抗氧化活性成分提取工艺及其抗氧化活性研究%Response surface optimization of the extraction process of Forsythia suspensa antioxidant active components and its antioxidant activity study

连翘 抗氧化活性 响应面分析 提取工艺
本研究旨在优化连翘抗氧化活性成分的超声辅助提取工艺,并对其提取物进行定性分析与抗氧化活性评价.采用单因素试验结合响应面法,以总抗氧化活性为评价指标,对连翘抗氧化活性成分超声辅助提取工艺进行优化;利用高效液相色谱(HPLC)对其提取物进行定性分析;通过对DPPH、ABTS+自由基的清除率以及Fe3+还原能力的测定,评估提取物的抗氧化活性.结果表明:优化后的连翘抗氧化活性成分超声辅助提取工艺条件为:乙醇浓度80%、料液比1∶30(g/mL)、超声温度60 ℃、超声时间50 min,此条件下,连翘抗氧化活性提取物的平均总抗氧化活性高达2.15 mmol/g,与理论预测值2.21 mmol/g相比较,其相对误差仅为2.71%.HPLC定性分析结果显示,提取物中含有连翘苷、连翘酯苷A、连翘酯素、芦丁、槲皮素、连翘酯苷I、松脂素-β-D-吡喃葡萄糖苷7种主要抗氧化活性物质.抗氧化活性评价表明,连翘抗氧化活性提取物具有较强清除DPPH、ABTS+自由基以及还原Fe3+的能力.本研究成功优化了连翘抗氧化活性成分的超声辅助提取工艺,该工艺稳定可行,且提取物富含多种抗氧化活性成分,展现出较强的抗氧化活性.%This study aimed to optimize the ultrasonic-assisted extraction process of antioxidant active components from Forsythia suspensa and to conduct qualitative analysis and antioxidant activity evaluation of the extracts.Using single factor experiments combined with response surface methodology,the ultrasonic-assisted extraction process of antioxidant active components from Forsythia suspensa was optimized with total antioxidant activity as the evaluation index.High-performance liquid chromatography(HPLC)was utilized for the qualitative analysis of the extracts.The antioxidant activity of the extracts was evaluated by the scavenging rates of DPPH and ABTS+radicals and the reducing power against Fe3+.The results showed that the optimal extraction conditions for the antioxidant active components from Forsythia suspensa were as follows:ethanol concentration of 80%,solid-liquid ratio of 1∶30(g/mL),ultrasonic temperature of 60℃,and ultrasonic time of 50 minutes.Under these conditions,the average total antioxidant activity of the extracts reached 2.15 mmol/g,with a relative error of only 2.71%compared with the theoretical predicted value of 2.21 mmol/g.The HPLC qualitative analysis results showed that the extracts contained seven main antioxidant active substances including forsythin,forsythin A,forsythin,rutin,quercetin,forsythin I,and pinoresinol-β-D-glucopyranoside.The antioxidant activity evaluation indicated that the extracts from Forsythia suspensa exhibited a strong ability to scavenge DPPH and ABTS+radicals,as well as to reduce Fe3+.This study successfully optimized the ultrasonic-assisted extraction process for the antioxidant active components from Forsythia suspensa,which was stable and feasible,and the extracts were rich in various antioxidant active components,showing strong antioxidant activity.This provided an important scientific basis for the further development and utilization of Forsythia suspensa resources.
