产纤溶酶菌株的分子鉴定及其液体产酶特点分析%STUDY ON TWO FIBRINOLYTIC ENZYME PRODUCING STRAINS IDENTIFICATION AND ITS LIQUID FERMENTATION CONDITIONS

纤溶酶 枯草芽孢杆菌 液体发酵 16S RRNA鉴定
通过对实验室分离筛选出的2株产纤溶酶能力较强的细菌菌株进行16S RRNA碱基序列测定,并与已发表的16S RRNA序列进行对比分析,确定2个菌株均属于枯草芽孢杆菌(BACILLUS SUBTILIS).在此基础上,对2个菌株液体发酵产纤溶酶的特点进行了对比分析.实验结果表明,2个菌株适宜的发酵产酶时间均为60~108 H.在优化的液体发酵条件下,2个菌株单位发酵液中纤溶酶平均产量可分别达到773.07 U/ML(菌株1)和962.28 U/ML(菌株2).%In this study,we identificated two excellent fibrinolytic enzyme producing strains from the traditional soybean diets with 16S rRNA gene;It was confirmed that two strains were Bacillus subtilis.Then we tested two strains ability of fibrinolytic enzyme producing in the liquid fermenter.The result confirmed that the suitable culture time of them were 60~108 h.At the suited liquid fermentation conditions,the highest fibrinolytic enzyme activity of strain 1 was 773.07 U/mL(strain1)and 962.28 U/mL(strain 2)respectively.
